| 内容記述 | To construct the restriction maps of the filamentous phage Xf and Xf2 DNA , the replicative form DNA （RF） was isolated from Xanthomonas campestris pv. oryzae cells infected with phage and digested with restriction endonucleases that recognize six bases. Xf-RF was cut by the treatment with restriction endonucleases BamHI, HindIII, SmaI and StuI at a respectively specific single site. Xf2-RF was cleaved by BamHI and XbaI at a single site, and PvuII and EcoRI at two sites. Based on the size and distribution patterns of the fragments generated by digestion with various combinations of these restriction endonucleases, restriction maps of the filamentous phage Xf-RF and Xf2-RF were constructed. | |||||

| 内容記述 | イネ白葉枯病菌繊維状ファージXfおよびXf2 DNAの切断点地図を作成する目的でファージ感染菌体から複製型環状二本鎖DNA （RF）を抽出し, 6塩基対の認識部位をもつ制限酵素で消化した。Xf-RFは制限酵素BamHI, HindIII, SmaIおよびStuIにより, それぞれ異なる1カ所で切断された。また, Xf2-RFはBamHIおよびXbalにより, それぞれ異なる1カ所で切断され, PvuIIおよびEcoRIにより2カ所で切断された。これらの制限酵素を種々組み合わせてXf-RFおよびXf2-RFを消化し, 生じた各断片の電気泳動パターンおよび各断片の分子量に基づき, Xf-RFおよびXf2-RFの制限酵素切断点地図を作成した。 | |||||
