| 内容記述 | We have earlier reported the devolopment of a high-performance liquid chromatography technique for quantitative determination of free tyrosine-O-sulfate (TyrS) in human urine with a sensitivity up to 0.5 pmol/analysis. Samples containing TyrS were chromatographed on a reverse-phase column using an ion-pair reagent containing tetrabutylammonium phosphate (TBA-P), and the TyrS eluted was determined based on the fluorescence displayed following post-column labeling with o-phthaldialdehyde. Using this procedure urine samples taken from fifty randomly selected healthy human subjects were analyzed, which revealed that the free TyrS level falls within the range of 6-88 μg/ml with a mean value of 28.42 ± 16.39 μg/ml. No sexual variation or age difference could be observed in the urinary tyrosine-O-sulfate content. | |||||

| 内容記述 | タンパク質の翻訳後の修飾としてチロシン残基の0-硫酸化は,タンパク質の機能制御または分泌のために重要であると考えられている.タンパク質に含まれる硫酸化チロシンの分析法としては,今まで35S放射線ラベル法を用いての検出法が行われてきたが,in situのタンパク質中の硫酸化チロシンおよび異化代謝物のフリーの硫酸化チロシンを分析するには不適当と考えられ,各種の高速液体クロマトグラフィー(HPLC)による硫酸化チロシンの分析法の検討を行った. 逆相,陽イオン交換,陰イオン交換およびイオンペア逆相HPLCを検討し,検出法にUV吸収およびオルトフタルアルデヒドによるポストカラム法での蛍光発色法を試みた.その結果,イオンペア逆相HPLCが硫酸化チロシンの分析に最も適していることが判明し,検出法では蛍光発色法においてピコモルオーダーの検出感度が得られた.これにより硫酸化チロシンタンパク質の異化代謝物である尿中のフリーの硫酸化チロシンの定量を,健康ヒト55検体について行ったところ6-88μg/mlの値を示し,男女に関係なく平均28.42±16.39μg/mlの結果が得られた. |
