@article{oai:miyazaki-u.repo.nii.ac.jp:00003265, author = {村上, 隆之 and Murakami, Takayuki and 斎藤, 勇夫 and Saito, Isao and 芦沢, 広三 and Ashizawa, Hirozo}, issue = {1}, journal = {宮崎大学農学部研究報告, Bulletin of the Faculty of Agriculture, Miyazaki University}, month = {Sep}, note = {鶏の尾腺細胞を電顕的に観察した.白色レグホーン種の尾腺をs-Collidineで緩衝した1%オスミック酸液(pH 7.4)で2~3時間固定後,漸強エタノール列で脱水,Luft法よりEpon812に包埋した.超薄切片を作ったのち,酢酸ウランと硝酸鉛の重染色を行ない,日立HU-11 D-S型電子顕微鏡で観察した.その結果は次のとおりである. 腺胞の辺縁部には未熟な腺細胞が位置し, 表層部に行くにしたがって成熟の度を加え最表層の細胞はホロクリン分泌によって腺腔内へ離脱する. 腺胞辺縁部の未熟細胞は少量の細胞質と大型の核を有し, 細胞質には遊離のリボゾームが豊富で, s-ER, r-ER, ミトコンドリア, トノフィラメントも少量含まれるが, Golgi装置の発達は悪く,脂肪滴は全く含まれない. 腺細胞は成熟するにつれて細胞質とs-ER の増加がみられ,それとともに脂肪滴並びに,顆粒状の内部構造を有し限界膜に包まれた dense bodies が出現する. 脂肪滴は限界膜を有さず扁平なs-ER で部分的に囲まれ,しかもs-ER 腔と脂肪滴との間にしばしば連絡部が見られる.一方,このように細胞内に脂肪滴が出現してもGolgi装置は未発達のままであり,両者の間に特別な関係は見い出せない. したがって鶏の尾腺細胞における脂質の合成や脂肪滴の形成はもっぱらs-ER によって行なわれる可能性が強い. s-ER がよく発達し,多量の脂肪滴を含む表層部の成熟細胞は,やがて核濃縮と細胞小器官の変性を経て,遂には細胞崩壊片となって腺腔内へ離脱する., The uropygial glands of domestic fowls, White Leghorn, were observed by electron microscopy. The specimens were fixed in ice-coid 1% osmium tetroxide bufferd with s-Collidine at pH 7.4. After 2-3 hours of fixation, the tissue blocks were dehydrated in a series of graded ethanol and embedded in Epon 812 according to the method of Luft. Ultrathin sections for electron microscopy were stained with uranyl acetate and lead citrate, and were examined with a Hitachi HU-11 D-S electron microscope. The observations were summarized as follows: In each acinus, the most peripheral cells are the youngest, grow as they approached the center of the acinus and the superficial cells are excreted by means of holocrine secretion into the lumen of acinus. The peripheral immature cells are small and have relatively large nuclei. Numerous free ribosomes are scattered throughout the cytoplasm. A few s-ER, r-ER, mitochondria and tonofilaments are also contained but lipid droplets have not formed as yet. Golgi complexes are not well developed. With the progress of cell maturity, an increase in cytoplasm and s-ER is observed. Lipid droplets and dense bodies which have a smooth surfaced limiting membrane and assume a granular configuration appear in the cytoplasm. The lipid droplets do not have a limiting membrane but tend to partially surrounded by flattened s-ER. Moreover, the lumen of s-ER frequently seem to be continuous with the lipid droplets. Golgi complexes, on the other hand, are always poorly developed and have no correlation with lipid droplets during the course of lipid droplets formation. These findings suggest that in the chicken uropygial glands, the s-ER is the main site of lipid synthesis and its droplets formation. In the superficial mature cells which are filled with abundant lipid droplets and s-ER, karyopiknosis and degenerations of cellular organelles appear soon.
