{"created":"2023-05-15T09:59:01.479132+00:00","id":3101,"links":{},"metadata":{"_buckets":{"deposit":"942975ba-04bc-46e3-8fca-3945e544cd20"},"_deposit":{"created_by":5,"id":"3101","owner":"5","owners":[5],"pid":{"revision_id":0,"type":"depid","value":"3101"},"status":"published"},"_oai":{"id":"oai:miyazaki-u.repo.nii.ac.jp:00003101","sets":["72","72:40"]},"author_link":["16322"],"item_10007_description_13":{"attribute_name":"抄録","attribute_value_mlt":[{"subitem_description":"デクスメデトミジン(DEX)とオレキシン(ORX)系の相互作用について調べた。最初に記録ニューロンがORX-Aに反応することを確認するために膜電流固定モードで3分間コントロールを測定した後ORX-A(10^<-6>M)を1分間灌流し、反応の有無を確認する。反応を認めたら、膜電位がベースラインに回復後15分以上経てからORX-Aの脱分極作用に対するDEX(10^<-6>M)の作用を調べた。ORX-A(10^<-6>M)を1分間灌流した後にDEX(10^<-6>M)を灌流すると最初は脱分極反応を認めたがその後は過分極反応を認めた。過分極反応は濃度依存性(DEX:3×10^<-9>-6×10^<-6>M)を示した。DEXの過分極作用はα_2受容体拮抗薬であるヨヒンビン(10^<-5>M)およびG蛋白共役型カリウムチャネル拮抗薬であるtertiapinに拮抗された。このDEXの過分極作用は、G蛋白共役型カリウムチャネルの活性化によるものだった。次にORX拮抗薬のDEXの過分極作用に対する影響を調べた。ORX-1受容体拮抗薬であるSB-334867(10^<-5>M)あるいは人工脳脊髄液(コントロール)を5分間灌流してからDEX(10^<-6>M)を2分間灌流して反応を比較した。ORX-1受容体拮抗薬投与前投与群とコントロール群で差はなかった。細胞レベルでDEXとORX系で相互作用はないことが明らかになった。","subitem_description_language":"ja","subitem_description_type":"Abstract"},{"subitem_description":"We investigated interaction between dexmedetomidine (DEX) and orexin (ORX) system. First, we identified ORX neuron by observing depolarizing effect induced by ORX application. Next, we examined the effect of DEX (10^<-6>M) on depolarization induced by ORX-A (10^<-6>M). We bath-applied ORX-A (10^<-6>M) for one minute and applied DEX (10^<-6>M). Depolarization induced by ORX followed by hyperpolarization induced by DEX was observed. DEX hyperpolarized PVN neuron dose-dependently (3x10^<-9>〜6x 10^<-6>M). The effect of DEX was blocked by an α2-adrenoceptor antagonist, yohimbine (10^<-5>M) and a G-protein-coupled inwardly rectifying K^+ (GIRK) channel blocker, tertiapin. Next, we examined the effect of SB-334867, an ORX-antagonist on the hyperpolarization induced by DEX. We first bath-applied SB-334867 (10^<-5>M) or artificial cerebrospinal fluid (control) for 5 min and applied DEX (10^<-6> M) for 2 min. We compared SB-334867 group or control group. Significant difference was not observed between two groups.
