Giant vesicles (GV) of a phospholipid containing a photopolymerizable diacetylene group, 2-di(10, 12-tricosadiynoyl)-sn-glycero-3-phosphocholine (DTPC), were prepared by a film rehydration technique under the controlling conditions such as the concentration, the incubation temperature, and the time of the DTPC solutions. The prepared GV were irradiated at 254 nm to form the polymer GV. Analysis by a confocal laser scanning microscope showed that the polymer GV prepared under the optimized conditions were a hollow sphere with 3-5 μm in diameter and < 1.0 μm thickness which consisted of a fluorescent polydiacetylene moiety of DTPC.
